Overview

NI-0801 is a fully human immunoglobulin G1 (IgG1) monoclonal antibody that targets chemokine (C-X-C motif) ligand 10 (CXCL10), also known as IP-10. CXCL10 is involved in the recruitment of inflammatory T cells into tissues such as the liver and plays a role in autoimmune and inflammatory diseases. NI-0801 binds with high affinity to human CXCL10 but not to other related chemokines like CXCL9 or CXCL11. By neutralizing CXCL10, NI-0801 inhibits its interaction with the receptor CXCR3, thereby blocking downstream signaling events such as calcium flux and lymphocyte migration. The drug was developed for conditions including primary biliary cholangitis (PBC), acute lung injury, adult respiratory distress syndrome, and vitiligo. Clinical trials have shown pharmacologic activity but did not demonstrate significant therapeutic benefit in PBC patients[1][2][3][8].
