NI-1701 (also known as TG-1801) is a fully human bispecific monoclonal antibody that targets both CD47 and CD19. It is designed to selectively block the "do not eat me" signal mediated by CD47 specifically on CD19-positive B cells, thereby promoting their phagocytosis and elimination by immune effector cells. This selectivity reduces off-target effects such as hemotoxicity commonly associated with broader CD47 blockade. The molecule features a fully competent IgG1 Fc domain, enabling antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). Developed using kappa-lambda body technology, NI-1701 is being investigated primarily for B cell malignancies including chronic lymphocytic leukemia (CLL), acute lymphocytic leukemia (ALL), non-Hodgkin lymphoma subtypes, and other B-cell lymphomas[1][2][3][4][5][6].
