NI-2801 is a bispecific antibody targeting CD47 and HER2, developed by Lightchain Bioscience using its proprietary κλ body platform. It is designed to treat HER2-positive solid tumors by selectively blocking the 'don't eat me' signal (CD47) on tumor cells that overexpress HER2. The molecule features a high-affinity HER2-targeting arm and an optimized low-affinity CD47-binding arm, which minimizes off-target binding to CD47 on healthy cells like red blood cells and platelets. NI-2801 incorporates a fully competent IgG1 Fc-domain to facilitate antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP). It is currently in the preclinical stage of development.
