Overview

NI-3201 is a bispecific monoclonal antibody designed to simultaneously target the PD-L1 immune checkpoint and the CD28 co-stimulatory receptor. Developed using the κλ-body platform, the drug is engineered to provide localized T-cell activation within the tumor microenvironment. Its dual mechanism of action involves blocking the PD-L1/PD-1 inhibitory pathway while conditionally providing T-cell co-stimulation (signal 2) via CD28 only when the antibody is engaged with PD-L1-expressing tumor or immune cells. This approach aims to enhance antitumor efficacy and promote immunologic memory while minimizing the systemic toxicities typically associated with non-targeted CD28 agonism.
