Nicaraven is a small molecule hydroxyl radical scavenger with antivasospastic and neuroprotective effects. It is under investigation primarily for the treatment of cerebrovascular diseases, including cerebral stroke and subarachnoid hemorrhage. Nicaraven acts by scavenging hydroxyl radicals in vivo, reducing oxidative stress and inflammation. It has demonstrated high permeability through the blood-brain barrier and shows potential to reduce delayed ischemic neurological deficits following aneurysmal subarachnoid hemorrhage. Mechanistically, it modulates Poly [ADP-ribose] polymerase 1 (PARP1) activity and activates the AMP-dependent protein kinase (AMPK)/Sirtuin 1 (Sirt1) signaling pathway while suppressing nuclear factor kappa B (NF-κB) activation in inflammatory models[1][2][4][8].
