Overview

Niceritrol is a niacin (nicotinic acid) derivative used as a hypolipidemic agent. It is an ester of pentaerythritol and nicotinic acid, slowly hydrolyzed in the body to release nicotinic acid. Niceritrol acts primarily by lowering cholesterol and triglyceride levels in plasma, particularly in the VLDL and LDL fractions. Its mechanism involves partial inhibition of free fatty acid release from adipose tissue and increased lipoprotein lipase activity, leading to improved lipid profiles. Additionally, it has vasodilatory properties and can inhibit platelet aggregation both directly and indirectly via lipid-lowering effects. Niceritrol has been used for treating hyperlipidemias (dyslipidemia) and peripheral vascular diseases.
