Overview

Niclosamide is an oral small molecule anthelmintic drug primarily used to treat tapeworm infections, including those caused by broad or fish tapeworm, dwarf tapeworm, and beef tapeworm. It acts by killing adult worms on contact in the intestine, which are then passed in the stool. The main mechanism of action is believed to be uncoupling oxidative phosphorylation in parasite mitochondria and stimulating ATPase activity, leading to energy depletion and death of the worm[1][2][3]. Beyond its antiparasitic use, niclosamide has been investigated for a wide range of other indications such as cancer (by inhibiting multiple oncogenic pathways including Wnt/β-catenin, mTORC1, STAT3, NF-κB), bacterial and viral infections, metabolic diseases (type II diabetes, NASH), autoimmune conditions (rheumatoid arthritis), and more[4][6][8]. It was previously marketed under the brand name Niclocide but was withdrawn from some markets.
