Overview

Niclosamide ethanolamine (also known as niclosamide olamine or NEN) is the ethanolamine salt of the salicylanilide anthelmintic drug niclosamide. It functions primarily as a mitochondrial uncoupler, a process that increases the permeability of the inner mitochondrial membrane to protons, thereby dissipating the proton gradient and uncoupling oxidative phosphorylation from ATP synthesis. This metabolic disruption leads to the activation of AMP-activated protein kinase (AMPK) and the inhibition of several key oncogenic signaling pathways, including Wnt/β-catenin, mTORC1, and STAT3. While historically utilized as a molluscicide and for treating tapeworm infections, NEN is being investigated for its potential in oncology, particularly for its ability to antagonize the Warburg effect and inhibit metastasis in colon and pancreatic cancers. It is also being explored for applications in metabolic disorders such as type 2 diabetes and nonalcoholic steatohepatitis (NASH) due to its favorable safety profile and multi-targeted mechanism of action.
