Overview

Nicotinamide adenine dinucleotide (NAD) is a coenzyme found in all living cells. It exists in two forms, the oxidized form (NAD+) and the reduced form (NADH). NAD+ plays a central role in cellular metabolism by acting as an electron carrier in redox reactions, particularly within glycolysis, the tricarboxylic acid cycle, and oxidative phosphorylation. Beyond its metabolic functions, NAD+ serves as a substrate for enzymes involved in DNA repair (such as poly(ADP-ribose) polymerases), protein deacetylation (sirtuins), and cell signaling pathways including ADP-ribosylation. It is synthesized via de novo pathways from amino acids or through salvage pathways recycling precursors like nicotinamide. In mammals, most tissues rely on salvage synthesis to maintain adequate levels of this essential molecule[1][2][3][4].
