Overview

Nicotinamide-expanded natural killer cells, also known as GDA-201 or NAM-NK cells, are allogeneic natural killer (NK) cells expanded ex vivo in the presence of nicotinamide (NAM) and interleukin-15 (IL-15). NAM enhances NK cell stability, survival, cytotoxic activity, glucose metabolism, resilience under oxidative stress, and cytokine production. These cells demonstrate potent cytolytic and antineoplastic activities and are being developed as cell therapies, particularly in combination with monoclonal antibodies such as rituximab or elotuzumab. Mechanistically, NAM-NK cells exhibit enhanced persistence after adoptive transfer, improved homing to lymph nodes via increased CD62L expression, and have been further engineered with CRISPR-Cas9 technology for CD38 knockout and chimeric antigen receptor (CAR) expression to improve targeting of multiple myeloma[1][2][3][4][7].
