Nicotinamide mononucleotide (NMN) is a nucleotide derived from ribose and nicotinamide. It serves as a direct precursor to nicotinamide adenine dinucleotide (NAD+), an essential coenzyme involved in cellular metabolism and energy production. NMN supplementation has been shown in animal models to increase NAD+ levels, potentially mitigating aging-related disorders such as oxidative stress, DNA damage, neurodegeneration, and inflammatory responses[1][3][5]. In humans, NMN is absorbed via the small intestine and converted to NAD+, though the exact mechanism remains under investigation[5]. Clinical studies have demonstrated that oral NMN increases blood NAD concentrations and may improve insulin sensitivity, aerobic capacity, physical performance (e.g., six-minute walking test), and subjective health assessments with good safety profiles at doses up to 900 mg daily[2][8]. Mechanistically, NMN’s effects are linked to increased SIRT1 protein expression and activation of heme oxygenase-1 signaling pathways[6]. While marketed for anti-aging benefits by supplement companies due to its role in boosting NAD+, robust evidence for these claims in humans is still limited.
