A nicotinamide N-methyltransferase antisense oligonucleotide is a therapeutic agent designed to inhibit the enzyme nicotinamide N-methyltransferase (NNMT). NNMT plays a crucial role in metabolism by methylating nicotinamide, a precursor to NAD+. Inhibition of NNMT leads to increased levels of S-adenosylmethionine (SAM) and NAD+ in adipose tissue, and upregulates the activity and expression of ornithine decarboxylase (ODC) and spermidine-spermine N(1)-acetyltransferase (SSAT). This results in increased polyamine flux and enhanced oxygen consumption in adipocytes. Furthermore, NNMT inhibition influences histone H3 lysine 4 methylation. This mechanism suggests its potential as a treatment for obesity and type 2 diabetes by augmenting cellular energy expenditure.
