Overview

Nicotinamide riboside + zinc is an investigational combination therapy consisting of two dietary supplements being evaluated by Cedars-Sinai Medical Center for the treatment of idiopathic pulmonary fibrosis (IPF). The therapy targets the ZIP8/SIRT1/NAD+ pathway to promote lung tissue regeneration and mitigate scarring. Nicotinamide riboside, a form of vitamin B3, serves as a precursor to nicotinamide adenine dinucleotide (NAD+), which is essential for DNA repair and mitochondrial function. Zinc is included to restore the function of ZIP8, a zinc transporter that is often downregulated in IPF, thereby aiding the regenerative potential of type 2 alveolar epithelial cells (lung stem cells). This combination is currently being studied in a decentralized Phase 2 clinical trial (NCT06567717) to assess its feasibility and efficacy in improving lung function and symptoms.
