Nicotinic acid mononucleotide (NaMN) is a crucial intermediate in the biosynthesis of nicotinamide adenine dinucleotide (NAD+), a pivotal cofactor in cellular metabolism. It is formed in the Preiss-Handler/de novo pathways of NAD+ synthesis and can also be generated from nicotinamide mononucleotide (NMN) through a base-exchange reaction mediated by the enzyme CD38. NaMN serves as a substrate for pyridine nucleotide adenylyltransferase (PNAT) enzymes, which catalyze its conversion to nicotinate adenine dinucleotide, an immediate precursor to NAD+. Its role in NAD+ metabolism makes it relevant for NAD+ replenishment strategies, with potential applications in nutritional and biomedical contexts.
