NID1-mRNA vaccine is a cutting-edge messenger RNA (mRNA) vaccine designed for the treatment and prevention of hepatocellular carcinoma (HCC), the most prevalent form of primary liver cancer. The vaccine incorporates full-length murine Nidogen-1 (NID1) mRNA into a novel lipid nanoparticle (LNP) formulation for delivery. Nidogen-1 is a glycoprotein prominently expressed in liver tumors and circulating small extracellular vesicles of HCC patients, playing a pivotal role in promoting HCC progression and metastasis. The NID1-mRNA vaccine works by inducing both cellular and humoral immune responses against NID1, thereby demonstrating significant therapeutic and preventive effects against HCC development. Preclinical studies in murine models have shown good tolerability and negligible systemic toxicity.
