Overview

Nifluridide (also known as L-27) is a small molecule tyrosine kinase inhibitor that selectively targets the keratinocyte growth factor receptor (KGFR, also known as FGFR2b). Originally developed by Eli Lilly as an ectoparasiticide and systemic insecticide, nifluridide was later identified through molecular modeling and investigated by researchers at the University of Oklahoma as a potent inhibitor of KGF-mediated breast cancer cell proliferation. In preclinical studies, nifluridide (L-27) demonstrated a dose-dependent reduction in the growth and metastasis of breast cancer xenografts with elevated expression of KGF, without showing signs of gross toxicity.
