Nifurpirinol is a synthetic nitrofuran derivative and small molecule antibiotic primarily used in veterinary medicine, especially for the treatment of bacterial infections in fish. It acts as a prodrug that requires activation by bacterial nitroreductase enzymes, leading to the formation of cytotoxic metabolites that cause cell death. Its mechanism of action involves inhibition of nucleic acid (particularly DNA) synthesis and interference with key bacterial enzyme systems involved in glucose and pyruvate metabolism, resulting in broad-spectrum antibacterial activity against various Gram-negative pathogens such as Vibrio anguillarum and Aeromonas species. Nifurpirinol is notable for its high potency at low concentrations compared to other nitrofuran antibiotics. While mainly used in aquaculture, it has also been explored as a research tool for targeted cell ablation using engineered nitroreductase-expressing models[1][4][5][6][7][8].
