Overview

NIK inhibitors are a class of small molecules designed to target and inhibit NF-κB-inducing kinase (NIK), also known as Mitogen-activated protein kinase kinase kinase 14 (MAP3K14). NIK is a central mediator of the non-canonical NF-κB signaling pathway, which regulates critical immune processes including B-cell maturation, lymphoid organogenesis, and the survival of various immune cells. Dysregulation or overactivation of NIK is implicated in several pathologies: in oncology, it contributes to tumor growth and the metabolic cross-talk driving cancer cachexia; in autoimmune diseases like rheumatoid arthritis, it promotes the expansion and differentiation of autoreactive B cells; and in respiratory conditions, it enhances the survival and activation of group 2 innate lymphoid cells (ILC2s) in severe asthma. Pharmaceutical development of NIK inhibitors, including programs explored by companies such as Janssen, aims to provide a novel therapeutic strategy for immune-mediated inflammatory disorders and various malignancies.
