NIK inhibitor 1 is a potent and selective small molecule inhibitor of NF-kappa-B-inducing kinase (NIK, also known as MAP3K14), with an IC50 of approximately 1 nM. Developed by Amgen, it serves as a pharmacological tool to investigate the non-canonical NF-kappa-B signaling pathway, which is frequently dysregulated in B-cell malignancies. The compound selectively targets NIK over other kinases and has demonstrated cytotoxic activity in multiple myeloma cell lines that exhibit NIK-dependent NF-kappa-B activation, often resulting from mutations in regulatory proteins like TRAF2, TRAF3, or BIRC2/3. Preclinical research suggests that NIK inhibitor 1 can synergize with IKKbeta inhibitors or glucocorticoids like dexamethasone to more effectively suppress tumor cell growth.
