Overview

Nilestriol is a synthetic estrogen, specifically the 3-cyclopentyl ether of ethinylestriol, making it a prodrug of ethinylestriol. It is described as a slowly-metabolized, long-acting estrogen and is more potent than its parent compound. Nilestriol acts as an estrogen receptor agonist and has been used primarily for hormone replacement therapy in postmenopausal women, including the prevention and treatment of postmenopausal osteoporosis. It was patented in 1971 and is marketed in China under the brand name Wei Ni An[1][2][3][4]. Nilestriol upregulates both ERα (estrogen receptor alpha) and ERβ (estrogen receptor beta) expression[2].
