NILK-3801 is a preclinical bispecific antibody candidate developed by LamKap Bio gamma, utilizing the proprietary κλ body (kappa-lambda body) platform. It is designed to treat Glypican-3 (GPC3)-positive solid tumors by providing a localized costimulatory signal to T cells. The molecule simultaneously binds to GPC3 on tumor cells and CD28 on T cells. This "Signal 2" bispecific approach is intended to complement "Signal 1" (CD3-based) T-cell engagers or endogenous TCR signaling, potentially enhancing T-cell activation and persistence within the tumor microenvironment while avoiding the systemic toxicity associated with non-targeted CD28 superagonists. By requiring GPC3 engagement for CD28 activation, NILK-3801 ensures that costimulation is restricted to the site of the tumor.
