Overview

Nilotinib is a small molecule tyrosine kinase inhibitor primarily used to treat Philadelphia chromosome positive chronic myeloid leukemia (Ph+ CML) in adults and children. It is indicated for newly diagnosed patients as well as those with chronic or accelerated phase Ph+ CML who are resistant or intolerant to prior therapy, including imatinib. Nilotinib works by selectively inhibiting the BCR-ABL tyrosine kinase, a fusion protein produced by the Philadelphia chromosome abnormality that drives leukemic cell proliferation. It also inhibits other kinases such as c-Kit and platelet-derived growth factor receptor (PDGFR). By blocking these kinases, nilotinib disrupts signaling pathways essential for cancer cell survival and proliferation[1][2][3].
