Overview

nilotinib + cytarabine + daunorubicin is a combination chemotherapy and targeted therapy regimen evaluated for the treatment of advanced BCR-ABL positive leukemias. The regimen consists of nilotinib, a second-generation tyrosine kinase inhibitor (TKI) that targets the BCR-ABL fusion protein, combined with standard '7+3' induction chemotherapy (cytarabine and daunorubicin). Nilotinib is approximately 20-fold more potent than imatinib in inhibiting the BCR-ABL protein. Cytarabine is an antimetabolite that inhibits DNA polymerase, while daunorubicin is an anthracycline that intercalates into DNA and inhibits topoisomerase II. This combination was investigated by Ulsan University Hospital as a potential bridging therapy to allogeneic hematopoietic cell transplantation for patients with myeloid blast crisis of chronic myeloid leukemia (CML-MBP) or BCR-ABL positive acute myeloid leukemia (AML). The clinical trial (Phase 2) was terminated early due to slow patient accrual.
