NIM-1001 is an orally active small molecule activator of Phosphatase and actin regulator 2 (PHACTR2) being developed for the treatment of systemic lupus erythematosus (SLE) and other autoimmune conditions. Developed by the NIMML Institute and NImmune Biotherapeutics, NIM-1001 is designed to enhance the differentiation, stability, and suppressive function of regulatory CD4+ T cells (Tregs). In preclinical models of SLE, such as MRL/lpr and NZB/W F1 mice, NIM-1001 treatment has demonstrated the ability to protect against kidney damage (proteinuria), increase splenic Treg populations, and reduce inflammatory cytokines and chemokines like CXCL1 and CCL4. By targeting the PHACTR2 pathway, the drug aims to restore immune tolerance in environments where Treg function is typically impaired.
