NIM-1324 is an oral, systemically distributed small-molecule therapeutic candidate that activates Lanthionine Synthetase C-like 2 (LANCL2), a surface membrane-associated receptor involved in modulating key cellular and molecular changes associated with autoimmune and inflammatory diseases. By activating the LANCL2 pathway, NIM-1324 increases the anti-inflammatory capacity and stability of regulatory CD4+ T cells and supports the metabolic demands of autophagy in phagocytes. In preclinical studies, it reduced production of interferon alpha in blood from systemic lupus erythematosus (SLE) patients and showed protective effects in mouse models of lupus, rheumatoid arthritis, multiple sclerosis, and other inflammatory indications. Phase 1 clinical trials demonstrated favorable safety, tolerability, dose-proportional pharmacokinetics up to 1500 mg daily with no severe adverse events or significant laboratory abnormalities[1][3][5]. It is being developed and manufactured by NImmune Biopharma. The primary indication is systemic lupus erythematosus, but preclinical data support potential for other autoimmune diseases such as rheumatoid arthritis and multiple sclerosis[1][3][6].
