Overview

Nimacimab is a first-in-class humanized monoclonal antibody developed by Skye Bioscience. It acts as a negative allosteric modulator of the cannabinoid CB1 receptor, selectively inhibiting CB1 signaling in the periphery while avoiding central nervous system (CNS) exposure. This mechanism aims to deliver anti-fibrotic, anti-inflammatory, and metabolic benefits without the neuropsychiatric side effects associated with earlier small molecule CB1 antagonists. Nimacimab is being investigated for obesity, chronic kidney disease, non-alcoholic steatohepatitis (NASH), diabetic gastroparesis, and diabetic nephropathies. Preclinical and early clinical studies have shown favorable safety profiles and promising efficacy signals in weight loss and metabolic parameters[2][5][6][7].
