Overview

Nimotuzumab is a humanized monoclonal antibody that targets the epidermal growth factor receptor (EGFR). By binding specifically to the extracellular domain of EGFR, it inhibits receptor activation and downstream signaling pathways involved in tumor cell proliferation, survival, and angiogenesis. This targeted action results in inhibition of tumor growth and induction of apoptosis in cancer cells overexpressing EGFR. Nimotuzumab also mediates immune responses such as antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC), activates natural killer cells, stimulates dendritic cell maturation, induces tumor-recognizing cytotoxic T-cells, and restores MHC-I expression on tumor cells. It is approved or used for squamous cell carcinoma of the head and neck (SCCHN), glioma/glioblastoma, nasopharyngeal cancer in various countries; it has orphan drug status for glioma in the US/EU.
