This combination therapy consists of nimotuzumab, a humanized IgG1 monoclonal antibody, and the TPF chemotherapy regimen, which includes docetaxel, cisplatin, and 5-fluorouracil. Nimotuzumab targets the extracellular domain of the epidermal growth factor receptor (EGFR), blocking ligand binding and inhibiting downstream signaling pathways involved in tumor cell proliferation, survival, and angiogenesis. The TPF components provide multi-targeted cytotoxic activity: docetaxel stabilizes microtubules to induce cell cycle arrest, cisplatin creates DNA cross-links that trigger apoptosis, and 5-fluorouracil inhibits thymidylate synthase to impair DNA replication. This specific regimen was evaluated in a Phase 2 clinical trial (NCT01425736) led by Wei Guo at Shanghai Ninth People's Hospital for patients with recurrent or metastatic head and neck squamous cell carcinoma (HNSCC).
