Nimotuzumab + painkiller refers to a therapeutic regimen combining the humanized monoclonal antibody Nimotuzumab with a structured analgesic protocol, typically following the WHO three-step pain ladder. Nimotuzumab, developed by Biotech Pharmaceutical Co., Ltd., is an IgG1 antibody that targets domain III of the extracellular region of the epidermal growth factor receptor (EGFR). It is characterized by an intermediate affinity for EGFR, which allows it to selectively bind to tumor cells with high EGFR expression while sparing healthy tissues with low EGFR density, thereby reducing common anti-EGFR toxicities such as severe cutaneous rash and hypomagnesemia. The analgesic component of the regimen includes a combination of non-opioids, weak opioids, and strong opioids (such as morphine or oxycodone) along with adjuvant analgesics to manage cancer-related pain. This combination is specifically being evaluated in locally advanced head and neck squamous cell carcinoma (HNSCC) to assess its impact on both tumor control and the quality of life through effective pain management during comprehensive treatment with chemoradiotherapy.
