Overview

This is a combination chemotherapy regimen consisting of three cytotoxic agents—nimustine, dacarbazine, and vincristine. Each drug has a distinct mechanism of action targeting cancer cells: - **Nimustine** is an alkylating agent from the nitrosourea class that crosslinks DNA strands, inhibiting DNA replication and transcription. - **Dacarbazine** is also an alkylating agent (triazene derivative) that works by methylating guanine bases in DNA, leading to inhibition of DNA synthesis and function. - **Vincristine** is a vinca alkaloid that binds to tubulin, inhibiting microtubule formation required for mitosis. This combination has been used in the treatment of various malignancies. There are published reports describing its use in regimens for high-grade glioma and other cancers. The combination exploits different mechanisms to maximize antitumor activity[2][5].
