Ninjurin1-targeting antisense oligonucleotides (ASOs) are a class of therapeutic agents designed to downregulate the expression of Ninjurin1 (Ninj1), a cell adhesion molecule involved in regulating inflammation and lytic cell death (pyroptosis). Developed by researchers at Seoul National University, Sookmyung Women’s University, and UMass Chan Medical School, these ASOs are being investigated for the treatment of Chronic Obstructive Pulmonary Disease (COPD), particularly cases induced by chronic inhalation of heavy metals like lead (Pb) and cadmium (Cd). By silencing Ninj1, particularly in pulmonary macrophages, these ASOs aim to mitigate COPD phenotypes such as mucus hypersecretion, alveolar airspace enlargement, and excessive inflammation.
