Overview

Nintedanib is an oral small-molecule multi-target receptor tyrosine kinase inhibitor with antifibrotic, antiangiogenic, and antitumor activity. It inhibits signaling through vascular endothelial growth factor receptors 1-3, fibroblast growth factor receptors 1-3, and platelet-derived growth factor receptors alpha and beta, which underlies its approved use in fibrosing interstitial lung diseases including idiopathic pulmonary fibrosis, systemic sclerosis-associated interstitial lung disease, and progressive fibrosing interstitial lung disease. The drug was developed by Boehringer Ingelheim and is marketed globally as Ofev; in China, Gyre Therapeutics acquired rights to a generic product marketed as Etorel. Beyond pulmonary fibrosis, nintedanib has been studied in multiple oncology indications, generally as a targeted antiangiogenic agent, but the provided clinical excerpts show limited single-agent activity in several tumor types.
