Overview

Nintedanib + carboplatin + paclitaxel is a combination regimen consisting of three drugs used primarily in the treatment of non-small cell lung cancer (NSCLC), including in patients with comorbid idiopathic pulmonary fibrosis (IPF), and has also been studied in advanced ovarian cancer. Nintedanib is an oral small molecule tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFRs), fibroblast growth factor receptors (FGFRs), and platelet-derived growth factor receptors (PDGFRs). By inhibiting these pathways, nintedanib exerts anti-angiogenic effects, suppressing tumor blood vessel formation and tumor progression. Carboplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking, leading to apoptosis of rapidly dividing cells. Paclitaxel is a microtubule-stabilizing agent that inhibits cell division by preventing microtubule depolymerization. This combination has been investigated for its efficacy and safety in NSCLC patients with IPF, where it may offer improved progression-free survival compared to chemotherapy alone but carries risks such as acute exacerbation of IPF[1][4][5]. In advanced ovarian cancer, the addition of nintedanib to standard carboplatin/paclitaxel did not demonstrate an overall survival benefit[6].
