Overview

Nintedanib + nivolumab is an investigational combination therapy for cancer, particularly advanced non-small cell lung cancer (NSCLC). Nintedanib is an oral small molecule tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR 1–3), fibroblast growth factor receptors (FGFR 1–3), and platelet-derived growth factor receptors (PDGFR α/β), thereby inhibiting angiogenesis, tumor proliferation, and the activity of immunosuppressive cancer-associated fibroblasts in the tumor microenvironment. Nivolumab is a monoclonal antibody immune checkpoint inhibitor targeting programmed death 1 receptor (PD-1) on T cells, enhancing anti-tumor immune responses. The rationale for combining these agents is to synergistically improve immunotherapy outcomes by modulating both the tumor vasculature/microenvironment and immune response. This combination has been evaluated in phase I/II clinical trials for advanced NSCLC[1][2][10].
