Overview

This combination therapy consists of nintedanib (BIBF 1120), a triple angiokinase inhibitor, administered in conjunction with the chemotherapy agents pegylated liposomal doxorubicin (PLD) and carboplatin (CBDCA). Nintedanib is a small molecule tyrosine kinase inhibitor that targets the vascular endothelial growth factor receptors (VEGFR 1-3), fibroblast growth factor receptors (FGFR 1-3), and platelet-derived growth factor receptors (PDGFR alpha and beta), which are essential for tumor angiogenesis and maintenance of the tumor microenvironment. Carboplatin is a platinum-based alkylating agent that induces DNA cross-linking, while pegylated liposomal doxorubicin is an anthracycline that inhibits topoisomerase II and intercalates into DNA. This specific regimen was developed by Boehringer Ingelheim and investigated in Phase I clinical trials for the treatment of patients with platinum-sensitive relapsed advanced epithelial ovarian cancer, fallopian tube cancer, or primary peritoneal cancer to determine the safety and tolerability of adding nintedanib to standard-of-care chemotherapy.
