Overview

NIO752 is an antisense oligonucleotide drug candidate developed by Novartis for the treatment of tauopathies, specifically progressive supranuclear palsy (PSP) and Alzheimer's disease. It works by targeting tau mRNA, thereby reducing the production of microtubule-associated protein tau in the brain. The reduction in tau levels is intended to slow or halt the formation of pathological tau aggregates that drive neurodegeneration in these diseases. NIO752 is administered via intrathecal injection to ensure delivery to the central nervous system. As of 2025, it is undergoing Phase 1 clinical trials for both PSP and Alzheimer's disease[1][2][3][4][5][6].
