NIOCH-14 is a chemically synthesized compound developed as an anti-smallpox medication. It is a derivative of tricyclodicarboxylic acid and an analog of tecovirimat (ST-246). The drug has demonstrated high activity against orthopoxviruses, including smallpox, monkeypox, cowpox, and ectromelia virus in various animal models[1][4]. When administered orally, NIOCH-14 is metabolized to its active form ST-246. It has shown good tissue availability for the lungs (100%), liver (69.6%), kidneys (63.3%), brain (26.8%), and spleen (20.3%), with an absolute bioavailability of 22.8% when administered orally to mice at a dose of 50 µg/g body weight[1][2]. The drug has completed Phase 1 clinical trials involving 90 healthy volunteers, demonstrating a favorable safety and tolerability profile[6]. It was licensed in the Russian Federation for the treatment of mpox (monkeypox)[5].
