NIP-121 is a **small molecule potassium channel opener** developed for cardiovascular indications, particularly as a vasodilator in the setting of **pulmonary hypertension** and systemic hypertension. It acts as an **activator of ATP-sensitive potassium (KATP) channels**, leading to membrane hyperpolarization and relaxation of vascular smooth muscle. In animal models, it has shown efficacy in reducing pulmonary and systemic vascular resistance and lowering blood pressure. The hypotensive and vasodilating effects can be blocked by KATP channel inhibitors (like glibenclamide), confirming the mechanism of action. NIP-121 is more effective than the calcium channel blocker nifedipine in reversing chronic hypoxia-induced pulmonary hypertension in rats. It does not operate through inhibition of Ca2+ release from the sarcoplasmic reticulum or protein kinase C activation.
