Overview

Niraparib + anlotinib is a combination therapy being investigated for the treatment of small cell lung cancer (SCLC), particularly in the maintenance setting or for relapsed/refractory cases. Niraparib is an orally active inhibitor of PARP-1 and PARP-2, which are enzymes involved in DNA repair; by inhibiting PARP, it induces synthetic lethality in cells with DNA repair deficiencies, especially those with homologous recombination deficiencies. Anlotinib is a multi-targeted tyrosine kinase inhibitor that suppresses tumor angiogenesis and growth by inhibiting VEGFR1-3, FGFR1-4, PDGFRα/β, and c-Kit. The combination aims to synergistically inhibit tumor progression by simultaneously targeting DNA repair mechanisms and the tumor microenvironment/angiogenesis. Clinical trials, such as those led by Jilin Province Cancer Hospital, explore this regimen's efficacy and safety in SCLC patients who have failed prior systemic therapies.
