Overview

A combination regimen of **niraparib** (a PARP inhibitor), **carboplatin** (a platinum-based chemotherapeutic agent), and **liposomal doxorubicin** (an anthracycline cytotoxic drug formulated for improved tolerability and efficacy), primarily investigated as therapy for platinum-sensitive and recurrent ovarian cancer. Niraparib inhibits poly(ADP-ribose) polymerases involved in DNA repair, enhancing tumor cell death, especially in homologous recombination-deficient tumors. Carboplatin induces DNA cross-linking and apoptosis. Liposomal doxorubicin intercalates DNA and inhibits topoisomerase II, blocking DNA replication in rapidly dividing cells. The combination is explored for potential synergistic cytotoxic and DNA repair–inhibitory effects in ovarian cancer[3][5].
