nirogacestat

Nirogacestat is an oral, selective small molecule gamma secretase inhibitor used to treat adults with desmoid tumors that cannot be removed by surgery or require systemic therapy. It works by blocking the activity of the gamma-secretase enzyme complex, which is responsible for proteolytic cleavage of multiple transmembrane proteins, including Notch receptors. Inhibition of Notch signaling disrupts cell growth and slows tumor progression. Nirogacestat has also been investigated in combination with B-cell maturation antigen (BCMA)-directed therapies for multiple myeloma, as it can increase membrane-bound BCMA density and potentially enhance the efficacy of these therapies[1][3][4][5][6][7][8].
