Overview

NIS793 + bevacizumab + chemotherapy is a combination regimen under investigation for second-line treatment in metastatic colorectal cancer (mCRC). NIS793 is a human IgG2 monoclonal antibody that specifically binds and inhibits transforming growth factor beta (TGF-β), a pathway implicated in tumor growth and resistance[2][4]. Bevacizumab is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis in tumors. The chemotherapy backbone includes modified FOLFOX6 (5-fluorouracil, leucovorin, oxaliplatin) or FOLFIRI (5-fluorouracil, leucovorin, irinotecan), determined by prior treatment. This combination aims to reverse resistance and enhance responsiveness to standard of care anti-cancer therapy[1][3]. NIS793 is developed by Novartis and is not yet approved; the regimen is being evaluated in phase II clinical studies.
