Overview

Nispomeben is an orally active, non‑opioid small‑molecule analgesic being developed by Novaremed for the treatment of chronic neuropathic pain, particularly painful diabetic peripheral neuropathy. It is a synthetic organic compound (alcohol/amide/phenolic benzene derivative) that exerts its effects as a modulator of the Src‑family tyrosine‑protein kinase Lyn, inhibiting Lyn kinase phosphorylation while showing no meaningful binding to classical pain targets such as opioid, serotonin, GABA, NMDA or cannabinoid receptors, or voltage‑gated sodium and calcium channels. Nispomeben has demonstrated clinically meaningful placebo‑corrected reductions in pain and favorable tolerability in a Phase 2a proof‑of‑concept trial in painful diabetic peripheral neuropathy, and is currently being evaluated in an NIH‑funded, randomized, placebo‑controlled Phase 2b trial; the program has received FDA Fast Track designation for painful diabetic peripheral neuropathy.[1][2][3][5][7][9][13][14]
