Nitro G

Nitro G is a sublingual spray formulation of nitroglycerin (glyceryl trinitrate), a potent vasodilator used for the acute relief and prophylaxis of angina pectoris. It is designed for rapid absorption through the oral mucosa, allowing the active ingredient to bypass first-pass hepatic metabolism and enter the systemic circulation quickly. Nitroglycerin acts as a prodrug that is bioactivated by mitochondrial aldehyde dehydrogenase (ALDH2) to release nitric oxide (NO). The released nitric oxide then stimulates soluble guanylate cyclase (sGC), increasing the intracellular concentration of cyclic guanosine monophosphate (cGMP). This increase in cGMP activates protein kinase G, leading to the dephosphorylation of myosin light chains and the relaxation of vascular smooth muscle. The resulting systemic vasodilation primarily affects the venous system, reducing venous return (preload) and myocardial wall tension, which decreases myocardial oxygen demand and alleviates anginal pain.
