Nitronazine (MN-08) is a small molecule nitrone derivative being developed by Guangzhou Magpie Pharmaceutical for the treatment of diabetic kidney disease (DKD) and ischemic stroke. Chemically, it is a hybrid molecule combining the structure of tetramethylpyrazine (TMP), an active component of the herb Ligusticum wallichii, with a nitrone moiety. This design allows it to function as a potent free radical scavenger and antioxidant. Nitronazine aims to mitigate oxidative stress and inflammatory damage, which are key drivers in the progression of diabetic nephropathy and neuronal injury following a stroke. It has been shown to activate the Nrf2 signaling pathway and inhibit the NLRP3 inflammasome. The drug is currently in Phase II clinical development in China for both DKD and acute ischemic stroke.
