Nivatrotamab is a fully humanized bispecific monoclonal antibody that targets both disialoganglioside GD2, which is highly expressed on certain tumor cells, and CD3 (T cell surface glycoprotein CD3) on T cells. By binding to these two targets, nivatrotamab cross-links T cells with GD2-expressing tumor cells, promoting a selective cytotoxic T lymphocyte response against the cancerous cells. This mechanism enables the immune system to recognize and destroy GD2-positive tumors such as neuroblastoma and osteosarcoma. Nivatrotamab is being developed by Y-mAbs Therapeutics in collaboration with Memorial Sloan Kettering Cancer Center for the treatment of relapsed or refractory neuroblastoma, osteosarcoma, small cell lung cancer, and other solid tumors expressing GD2. The drug has received orphan drug designation for neuroblastoma from the FDA and remains in clinical development[1][5][7].
