Overview

Nivisnebart is a human monoclonal antibody of the IgG1κ subtype that specifically targets sortilin (SORT1). The drug works by blocking sortilin activity, which is a protein that shuttles progranulin into cellular degradation pathways. By inhibiting sortilin, nivisnebart increases progranulin levels.[1][4] Progranulin serves as a key regulator of immune activity in the brain and is involved in neuroprotection. The drug was initially developed by Alector and is now being developed in collaboration with GSK for neurodegenerative diseases, particularly Alzheimer's disease.[4][5]
