Nivo813 is a dual-blocking anti-PD-1/ALK-1 bispecific antibody being developed by Kintor Pharmaceutical for the treatment of solid tumors. It is designed to simultaneously target Programmed Cell Death Protein 1 (PD-1) and Activin Receptor-Like Kinase 1 (ALK-1). By blocking PD-1, Nivo813 enhances T-cell-mediated anti-tumor immune responses, while its inhibition of ALK-1, a member of the TGF-beta superfamily, suppresses tumor angiogenesis. In preclinical studies, Nivo813 demonstrated high binding affinity to both cell surface PD-1 and ALK-1, dose-dependent anti-angiogenesis activity in vitro, and superior anti-tumor efficacy compared to nivolumab in vivo.
