Drug intelligence / Profile preview
Nivolumab + brentuximab vedotin is a **combination therapy** comprising two distinct drugs: nivolumab, an immune checkpoint inhibitor targeting the PD-1 receptor, and brentuximab vedotin, an antibody-drug conjugate (ADC) targeting CD30. Nivolumab is a recombinant human monoclonal antibody that blocks the programmed death-1 (PD-1) receptor, thereby enhancing T-cell immune responses against cancer cells. Brentuximab vedotin consists of an anti-CD30 monoclonal antibody attached via a protease-cleavable linker to the cytotoxic agent monomethyl auristatin E (MMAE); upon binding to CD30-expressing tumor cells, the conjugate is internalized, releasing MMAE to induce cell cycle arrest and apoptosis through microtubule disruption. This combination addresses complementary mechanisms in lymphoma: immune activation by nivolumab and targeted cytotoxicity by brentuximab vedotin. The regimen has been studied in relapsed/refractory and newly diagnosed classic Hodgkin lymphoma, both as consolidation post-autologous stem cell transplant and in frontline settings with or without additional chemotherapeutic agents[1][2][3][5][6].
