Overview

**nivolumab + gemcitabine + liposomal doxorubicin** is an investigational combination regimen consisting of a monoclonal antibody checkpoint inhibitor (nivolumab), a nucleoside analog antimetabolite (gemcitabine), and a pegylated liposomal formulation of an anthracycline antibiotic (doxorubicin). Nivolumab is a human IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), restoring antitumor immune response. Gemcitabine inhibits DNA synthesis, leading to apoptosis of dividing tumor cells. Liposomal doxorubicin intercalates DNA and inhibits topoisomerase II, also producing free radicals that damage cellular components. The combination aims to synergize immune checkpoint inhibition (immune activation) with cytotoxic chemotherapy, mainly explored in relapsed/refractory malignancies, including classical Hodgkin lymphoma and platinum-resistant ovarian cancer. As of 2025, there is no unique brand name or regulatory approval for the three-drug combination, and use is limited to clinical study settings.
